DEEP-well microfluidics for arrayed colorimetric LAMP analysis

Abstract

This paper presents a microfluidic device for colorimetric loop-mediated isothermal amplification (LAMP) detection based on hydroxynapthol blue (HNB), long optical pathlength reaction wells, and two-phase liquid loading into a novel array geometry. We show that an inexpensive camera can be used with diffuse illumination to detect colorimetric changes due to LAMP reactions in sub-microliter wells.
