Application of displacement thin-layer chromatography to toad-poison bufadienolides

Abstract

We have investigated displacement thin-layer chromatography with organic bases N,N-dimethyl-1,3-propanediamine, N,N-dimethylcyclohexylamine, and triethylamine for the separation of seven toad-poison bufadienolides - resibufogenin, cinobufagin, bufalin, bufotalin, cinobufotalin, telocinobufagin, and gamabufotalin. The multiple use of the mobile phase mixtures n-hexane-acetone, n-hexane-ethyl acetate, n-hexane-dichloromethane- acetone, n-hexane-dichloromethane-methanol, and n-hexane-ethyl acetate- methanol, each containing 3% (v/v) organic bases, furnished effective separation of the bufadienolides. The characteristics of the separations and the visualization of the bufadienolides by derivatization are also discussed. The technique has been applied to the analysis of the major bufadienolides in the Chinese traditional drug Ch'an Su.
