The accelerating tuberculosis (TB) epidemic in settings where HIV is highly prevalent demands new tools for TB control. One are of great need, especially for pediatric TB, is improved diagnosis. The standard approaches of sputum smear microscopy and culture are insensitive, particularly in children, as well as in HIV-infected persons of all ages, and require laboratory resources and skilled personnel. In an effort to identify antigens of Mycobacterium tuberculosis (Mtb) that can be used to develop new antibody-based tests for the diagnosis of TB in children, a serum screen of TB infected children on a TB protein microarray is planned. Harvard Institute of Proteomics, under the directorship of Dr. Joshua LaBaer, will manufacture the TB protein microarrays. The time line for the work to be completed is as follows: Year 1: Transfer the second set of 1500 TB clones into NAPPA expression vectors. A small number of these expression clones will then be sent for sequencing as a quality control check. These clones are then printed in sets of 90 arrays each with180 assays delivered to Children's Hospital, Boston. Year 2: Another 2 sets of 180 slides for each 1500 spot array will be printed and delivered to Children's Hospital, Boston.
