Autoantibody Biomarker Discovery in Inflammatory Bowel Diseases Autoantibody Biomarker Discovery in Inflammatory Bowel Disease Using Immunoproteomics ABSTRACT: Patients with inflammatory bowel disease (IBD) have identifiable antibodies against microbial and human proteins that can serve as potential serological biomarkers in the management of these patients. The primary aim of this study is to identify specific antibodies against human proteins in patients with ulcerative colitis (UC) and Crohns disease (CD). The secondary aims are to determine if the identified antibody profiles can differentiate IBD from healthy controls and differentiate between UC and CD. The antibodies will be identified on an innovative protein microarray platform, nucleic acid programmable protein array (NAPPA), developed by Dr. LaBaer at ASU. We have conducted a pilot project with 30 serum samples to demonstrate the feasibility of detecting specific autoantibodies for inflammatory bowel disease (IBD) using Nucleic Acid Programmable Protein Arrays (NAPPA). The Mayo ASU seed grant will expand that study to include the screening of 150 patients against 2000 human genes in order to obtain statistical data. The preliminary data generated from this study will be used to apply for a NIH R01 grant to conduct a full-scale statistically-powered three phase study (pre-screening, discovery and validation) against all ~12,000 human genes in our collection. The SHARE registry (Bioinformatics, Giesel School of Medicine, Dartmouth) will be utilized for collection, storage and analysis of data.
